Introduction

The present invention relates to the production of an unerodible, biocompatible, transparent and sterilizable hydrogel scaffold for a 3D cell culture which can be used to culture a variety of different cells for prolonged periods of time and potentially in vivo for regenerative tissue procedures.

Technical features

The scaffold is based on a polyacrylic acid (FDA approved biocompatible material). It is able to resist to sterilization by moist heat process in autoclave and to freeze drying. Its highly interconnected porous structure is suitable for cell migration and diffusion of nutrients or metabolic wastes. It is transparent, thus allowing the microscopy visual monitoring of cell wellness and morphology. Suitable for routine cell viability tests (MTT, MTS, NRU, Alamar Blue). Easy to use, as it shows enough mechanical strength to be handled during cell culturing and assays. Easily scalable and economic. Potential biocompatibility and the its cross-linked structure make it potentially applicable for devices for tissue regeneration or advanced therapy. Moreover the carboxylic acid functionality provides a useful linkage for subsequent surface modifications and bio-conjugation.
